Lo decide papa Paolo IV Carafa nel 1555 e la segregazione sopravvive per tre secoli, a dispetto degli stravolgimenti della storia, a dispetto della popolazione che continua a crescere fino ad accogliere tutti gli ebrei dello Stato Pontificio, deportati a Roma. Non si poteva uscire dal ghetto di notte, non si potevano svolgere lavori comuni.
